22.5 inch semi-truck wheels

I havea 2004 Dodge Ram Quad Cab Longbed 5.9 diesel dually (305/555) six-speed w/ anti-slip diff. The engine and suspension are completely stock. The wheels/tires are 235/80-17 LT. I want to run 22.5 x 8 wheels (8 to 10 stud adapters) w/ 275/80-22.5 tires. How will this effect my gas mileage and speedometer?

Well, you want to look at the overall diameter of the rolling stock. That will probably be the biggest factor in whether or not it will affect your mileage. Also, unsprung weight and rotational mass will be a factor. In terms of the speedometer correction, you should be able to fix that with a plug-in module from a company like Hypertech or Superchips.
